MC‐RSCs were treated with Crx or Nrl siRNA or GSK3 inhibitor SB‐216763, qRT‐PCR and Western blot analysis showed that si‐Crx significantly reduced Crx and Nrl mRNA and protein levels, si‐Nrl significantly reduced Nrl mRNA and protein levels, while SB‐216763increased nuclear β‐catenin/total β‐catenin ratio. The positive rate ofphotoreceptor‐like cells in Crx or Nrl siRNA group was20.43 ± 6.21% and 22.56 ± 5.31%, respectively, significantly lowerthan NC siRNA group (42.31 ± 7.69%) (F = 26.99, P < 0.05). In contrast, the positive rate of photoreceptor‐like cells in SB‐216763 treated group (23.42 ± 6.44%) was significantly lower than vehicletreated group (41.31 ± 7.58%) (t=3.593, P < 0.05) (Figure 3A‐C).
