Therefore, we further investigated the PRDX1 protein level innuclear and cytosol of ACHN cell. Along with increasing totalPRDX1, obvious translocation of PRDX1 toward the nucleus was also observed, indicated that PA/GPA not only raised theexpression of mRNA and protein of PRDX1 but also forced itinto the nucleus (Figure 3D,E). Some studies have shown thatPRDX1 in the nucleus leads to inhibition of NF-κB activation,a typical oncogene involved in most cancers.20 These datafurther suggested that the protein level of PRDX1, but notantioxidant activity, is critical in regulating the ROS level andthe inactivation of NF-κB.
