The factors necessary for the growth of Lactobacillus brevis include calcium pantothenate, niacin, thiamine (VB1), and folic acid, but riboflavin (VB2), pyridoxal (VB6), and VB12 are not required. The properties of the NAD enzyme of Lactobacillus brevis are shown in Table 2. According to different strain sources, the optimal pH of NAD enzyme is between 4.0-5.2, the optimal temperature is between 30-45°C, and the molecular weight is between 53-62kDa. Among them, the NAD activity of strain IFO12005 can be activated by NH4+ (Kazumi HiRAGA et al. 2008). The enzyme activity of NAD enzyme decreases sharply when the pH is close to neutral or the temperature is close to 50°C. Studies have shown that after the carbon end of NAD is truncated, the range of action of NAD enzyme can be close to the neutral pH condition (Kai Yu et al. 2012 ). Fang Hui et al. screened the NAD enzyme after G364P mutation, and its thermal stability was significantly enhanced.
