Galactose is present in various polymers and glycoproteins in cell walls, including β-1,4-galactan and xyloglucan. To determine if the increase in D-Gal was related to increased β-1,4-galactan, cell wall material was digested with endo-β-1,4-galactanase from Aspergillius niger , and the D-Gal content was analyzed in the solubilized and residual materials. Treatment with endo-β-1,4-galactanase released more D-Gal from the cell wall of salt-treated plants than from control plants, whereas the residual material did not show any difference (Figure 1B). This indicate that the increase in total D-Gal is specifically due to an increase in pectic β-1,4-galactan content.
