Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) is a method for detecting the effect of protein separation. This method is based on the difference in molecular weight of protein subunits to achieve protein separation. In this experiment, recombinant human follicle stimulating hormone (rhFSH) was used as a sample, and the separation of the gel and protein bands were studied by SDS-PAGE electrophoresis. The gel separation effect of different SDS content is different, and the protein bands detected by different loading volumes are also different. In this experiment, the separation effect of 8% and 13.5% gels was first compared, and it was concluded that the 13.5% gel was suitable for the detection of rhFSH protein. Change the sample loading volume, use the selected 13.5% gel for electrophoresis detection, and compare and analyze the gel map to find that different loading volumes have different effects on the gel band, which is suitable for rhFSH sample loading volume of 10 μL. Finally, the protein activity of rhFSH was studied, and the pH and temperature were tested separately. Different pH values were selected to accelerate the reaction in a 30 ° C incubator. When the pH value is 7, rhFSH protein activity is relatively stable.
